remove this file C:\Program Files (x86)\JDownloader 2\jd\plugins\hoster\IFileIt.class
and let the update replace/install it again
seems something blocked access to the file
make sure no java processes are running, then start Updater.jar

I try to update because Im having problem with Mediafire.

The updater.jar worked, but only with the file, doesn't work with the program's updater button.

But still failing the Mediafire.com (plugin crash) downloads after the update.

I don't know what to do.

If you have a problem with mediafire, please open a new thread in the Hoster Plugin Forum!

Nicht genug Rechte um nach C:\Program Files (x86)\
seems you started jd with wrong rights!
this message means: not enough rights to move file

you have rights missmatch!
you have to start jd as same user as you installed it with!
rightclick on the jd folder, properties
security tab, edit
and give every user full control!

about mediafire, please do as pspzocker said
